The Rapid Fluorescent Focus Inhibition Testing is an in vitro neutralization testing for rabies virus. It measures the neutralizing activity (potency so to speak) of either human sera or in our case human recombinant antibodies.
It involves incubating the rabies virus with antibody to allow neutralization followed by addition of mouse neuroblastoma cells. The virus/mAb/cell mixture is subsequently incubated for 24h at 37C after which the cells are fixed with acetone and stained with an anti-rabies FITC labeled mAb to visualize infected cells.
Infected cells are counted and benchmarked against a reference to determine the potency.
