NTA for Particle Concentrations and Western Blot Analysis ( TET3, CD81, CD 63, and CD9)
Project Description
Contract Laboratory needed for plant NTA for particle concentrations and Western Blot analysis for TET3, CD81, CD 63, and CD9.
Two sets of samples will be provided. One set will be chilled and will requiring starting with step 1A. The other set will be frozen and will start with step 1B.
1A. For non-frozen samples centrifuge using Amicon Ultra-4 centrifugal filter units. Freeze those until ready for step 1B (or begin Step 1B with freshly centrifuged samples).
1B. Samples will need to be thawed. Do not thaw in a heat bath, if possible only thaw in the refrigerator.
2. Centrifuge samples at 10,000-12,000 xg (depending on rpm conversion for centrifuge) for 10 minutes, and remove supernatant for analysis.
Analysis:
1. Perform NTA for particle concentrations and average size/size distribution.
2. Perform Western Blot analysis for TET3, CD81, CD 63, and CD9.
Samples should be kept on ice at all times possible during final processing and analysis steps.
