Canada ISO bioanalytical laboratory needed forbioanalytical research study for Evaluation of the effect of crude oil contaminants as in-vitro endocrine disruptor and cell proliferation
This research will evaluates the potential endocrine disruptor activity of substances by measuring whether and to what extent a substance induces cell proliferation via estrogen receptor (ER), androgen receptor (AR) (PR) dependent pathways. This research is aimed at screening of test samples to quantify functional activities, either agonist or antagonist, that they may exert against the human androgen receptor and estrogen (ER?) estrogen (ER?).
We shall assay for the following:
1. Agonist dose-response of the AR Reporter Assay
2. Antagonist dose-response of the AR reporter Assay
3. Human ER? and ER? Agonist assays
4. Human ER? antagonist
5. Human ER? antagonist Assay
the statistical analysis shall be carried out as outline below.
Average relative light units (RLU) and corresponding standard deviation (SD) values will be determined for each model concentration (n ? 6). Signal-to-background (S/B) and Z? values will be calculated using Hill?s equation as described by Zhang, et al. (1999a). Non-linear regression and regression analysis, inhibitory concentration (IC50) as well as (Ki) and effective concentration (EC50) analysis will be performed using GraphPad Prism version 6 software. Low percentage coefficient of variation (% CV), high S/B and Z scores will confirm the robust performance of this Assay.
