Quantifying Acrylamide in Heat-Processed Foods

MULTIPLE TESTING
Analytical laboratory (the Philippines preferred) needed to quantify acrylamide in commonly consumed heat-processed and carbohydrate-rich foods using the GC-MS/MS detection method. We'd also like the lab to extract acrylamide from various food samples, following a modified protocol (Jezussek & Schieberle, 2003), which we will implement in our analysis.
Methodology:
1. Sample Collection: We plan to collect commonly consumed heat-processed and carbohydrate-rich foods in the Philippines, which will be vacuum-packed and stored at -80°C until analysis.
2. Chemicals and Reagents: The chemicals involved include acrylamide (50 μg/mL), [13C3]-acrylamide as an internal standard, and other reagents such as acetonitrile/H2O (85:15), n-hexane, ethyl acetate, and sodium sulfate.
3. Acrylamide Extraction: The extraction process includes homogenization with acetonitrile/H2O, sonication, liquid-liquid extraction with n-hexane, and application on an Extrelut NT 20 cartridge for elution with ethyl acetate.
4. Standard Preparation: Calibration standards for quantification are prepared using [13C3]-acrylamide stock solution, and acrylamide stock solution, with concentrations ranging from 0.023 to 2.0 μg/mL.
5. Acrylamide Quantification: Quantification will be done using an Agilent 8890 GC system coupled with an Agilent 7000D triple quadrupole mass spectrometer, employing multiple reaction monitoring for detection.
6. Method Validation: We will validate the method in terms of precision, accuracy, and sensitivity, ensuring that %RSD values remain below 20% and LoD and LoQ are set at 2.10 μg/kg and 7.60 μg/kg, respectively.
